The new Unity development will see the old Rumford Court building replaced by a £60m luxury flat scheme. Project leaders say they expect the road closure to last a week.
Construction teams will tonight bring in a crane to begin demolition work, which is expected to take 20 weeks.
The 27- storey development, which will be a mixture of commercial space and flats in two towers, will be completed by 2006.
A spokesman for Rumford Investments said: "Obviously, with any development of this high quality there will be some disruptions.
"Whatever the short-term problems, in the long term this building will be befitting to Liverpool as it comes up to Capital of Culture.
"This is yet another example of Liverpool's ambition.
